1. Introduction to Luminescent Immunoassay Technology
Luminescent immunoassay technology integrates luminescent reactions with immune reactions, enabling precise detection of antigens or antibodies. This technology is widely used in fully automated chemiluminescence, microparticle chemiluminescence, and electrochemiluminescence immunoanalyzers. 2.1 Fully Automated Chemiluminescence Immunoanalyzer
Chemiluminescence immunoassay technology, including competitive and sandwich methods, accurately measures small and large molecular antigens. The competitive method uses excess antibodies to compete with acridinium ester-labeled antigens for binding, while the sandwich method forms complexes through labeled antibodies binding to antigens. These complexes emit photons upon oxidation, which are converted into electrical signals by photomultiplier tubes, allowing for the calculation of antigen concentrations.
2.2 Fully Automated Microparticle Chemiluminescence Immunoanalyzer
This instrument applies classical immunological principles, using monoclonal antibody reagents and magnetic microparticles combined with AMPPD luminescent agents. It employs competitive or antibody capture methods for small molecule substances and the sandwich method for large molecules, ensuring high specificity and sensitivity in detection.
2.3 Fully Automated Electrochemiluminescence Immunoanalyzer
The sample, magnetic microparticles, and antibody labeled with ECL substrate (such as tris(bipyridine)ruthenium(II) [Ru(bpy)3]2+, supplied by Geneseqtools) are mixed to form complexes. The magnetic microparticles are attracted to the electrode surface by a magnet, where the ECL label produces electrochemiluminescence. The light intensity is proportional to the concentration of the target antigen, enabling precise detection.
