1. About α-L-fucosidase

α-L-fucosidase (AFU) is a lysosomal acid hydrolase widely found in various tissues, blood, and bodily fluids of the human body. Its primary function is to catalyze the hydrolysis of large molecules such as glycoproteins and glycolipids containing fucose residues, thus participating in complex metabolic processes. 

AFU not only plays an essential role under normal physiological conditions but also exhibits unique diagnostic value in various diseases. Particularly noteworthy is its role in the early screening and diagnosis of liver cancer. Studies have shown that AFU has a sensitivity and specificity of 75% and 90%, respectively, in diagnosing liver cancer, higher than traditional tumor markers like alpha-fetoprotein (AFP). Moreover, combined detection of AFU and AFP can increase the sensitivity of diagnosing primary liver cancer to 91.30%, with an accuracy rate reaching 91.04%, significantly reducing the likelihood of missed diagnoses and enhancing diagnostic precision and comprehensiveness.




2. Importance of Enzyme Substrates and Detection Principles

α-L-fucosidase operates like a specialized task force dealing with L-fucose-containing complex molecules in the body. These molecules are akin to encrypted information that can only be unlocked and decomposed by specific keys—enzyme substrates. The choice of substrate is critical for ensuring accurate detection results. An ideal substrate should possess high specificity, rapid reaction, stability, and ease of use. Detection Principle: 2-Chloro-4-nitrophenyl-α-L-fucopyranoside (CNP-AFU) is hydrolyzed by α-L-fucosidase in the sample to produce 2-chloro-4-nitrophenol (CNP). Measuring the increase in absorbance of CNP allows for determining AFU activity.
