I. Working Principle

In Sequencing By Synthesis (SBS) technology, the identification of each base relies on a key design: reversible termination and cleavable fluorescent labeling.

Its chemical core lies in — the azido group (-N₃) serving as the linker.

3ʹ-blocked, fluorescently labeled dUTP

The working principle is as follows:

1. Precise Pairing: Four types of 3ʹ-blocked dNTPs are labeled with fluorophores of different wavelengths. Under the action of DNA polymerase, these fluorescently labeled dNTPs precisely pair with the bases on the sample DNA single strand, extending the complementary strand.

2. Fluorescence Detection: Because the 3ʹ-position of the dNTP is blocked, the DNA chain can only add one base at a time. The sequencer determines which base (A, T, C, G) was incorporated by detecting the fluorescence wavelength in each reaction cycle.

3. Cleavage and Regeneration: After detection, a trivalent phosphine reagent (PR₃) specifically cleaves the azido linker connecting the fluorophore and the dNTP, washing away the fluorophore and simultaneously removing the 3ʹ-blocking group, restoring the DNA chain's extension capability for the next cycle.
