S-2732 is a pNA-based chromogenic substrate, consisting of the synthetic peptide Suc-Ile-Glu(γ-pip)-Gly-Arg coupled with the pNA chromophore via an amide bond. Its core detection logic can be summarized in three stages: "cleavage – release – color development":

1. Enzyme-specific recognition and cleavage

The peptide sequence of S-2732 (Ile-Glu-γ-pip-Gly-Arg) is a specific recognition site for coagulation Factor Xa. FXa is upstream of thrombin in the coagulation cascade and is the core target of heparin's anticoagulant action. When active FXa is present in the system, the enzyme specifically recognizes and cleaves the Arg-pNA bond in the substrate, which is the classical recognition sequence for FXa cleavage.

2. Release of the pNA chromophore

In the intact substrate, pNA is covalently linked to the peptide and does not produce a significant color signal in the un-released state. After cleavage, pNA is released as free p-nitroaniline molecules.

3. Production and quantification of yellow color signal

Free pNA appears yellow in solution, exhibiting a characteristic absorption peak at 405 nm. Absorbance is proportional to residual FXa activity and inversely proportional to heparin activity in the sample.
